Bolt defeated fellow Jamaican Yohan Blake and American star Tyson Gay, who took the bronze, at Olympic Stadium in London on Sunday.
Bolt now joins American track legend Carl Lewis (1984, 1988) as the only men to win back-to-back Olympic gold in the 100 meters.
Bolt trailed in the early seconds of Sunday's race, running in fifth place at the halfway mark. But the Jamaican surged ahead to a decisive victory, raising a triumphant finger in the air as he crossed the finish line.
He then ran a victory lap around the track, high-fiving fans, kissing the track and even flipping a somersault.
Bolt's compatriot Blake took the silver, finishing in 9.75 seconds. Gay came in third, in 9.79 seconds.
Bolt became an international superstar after he bolted to the gold in the 100 meters, 200 meters and 4x100 meter relay at the 2008 Beijing Games. He set Olympic and world records in both the 100 and 200, running 9.69 second in the former and 19.30 in the latter.
